1. Ionic imbalance within the muscle: Muscle contraction requires Ca1 2 3 4 5+ ions to interact with troponin for exposing the actin-binding site to myosin head.

If deficiency of Ca2+ occurs in the body, then muscles do not get the required Ca2+ ions for contraction.

2. Nervous fatigue and loss of desire: The contraction of a muscle is controlled by nerves. In central fatigue or “psy­chological fatigue,” brain feels tired. This leads to fatigue.

3. Metabolic fatigue: Depletion of ATP or glycogen results in fatigue due to unavailability of energy. Metabolites like Mg2+ ions induce fatigue by inhibiting the release of Ca2+ ions or reducing sensitivity of troponin to Ca2+ ions.

4. Exercise and aging: With aging of animal, the levels of ATP, CTP, and myoglobin decrease and it reduces the muscle’s ability to function. Training and exercise increase the metabolic capacity of a muscle, which delays the onset of muscle fatigue.

5. Lactic acid accumulation: Lactic acid is the by-product of anaerobic respiration, which strongly contributes to mus­cle fatigue.
